The UniSep C8 columns feature ultra-pure spherical silica, bonded to an octyl silane which contains an ether linkage close to the silica surface. Since the ether group is polar, water can easily penetrate and hydrate the silica surface, allowing the analyte greater access to the binding sites.
The advantage of using these columns lies in the flexibility gained when developing a method for highly water-soluble compounds. With traditional C8 columns, vitamins and polar compounds with low molecular weights require the addition of an ion-pairing reagent to the mobile phase to facilitate binding. These reagents can interfere with the peak identification.
