Kits are designed for rabbit or mouse primary antibodies and include reagents and buffers necessary for performing 10 IP-Western blots to detect proteins from immunoprecipitation. One-Step IP-Western immuno-detection technology reduces total analysis time for Western or dot blot to one hour and eliminates contamination from heavy and light chains, as well as proteins A and G. Applications include protein expression and gene function studies, examinations of alternative splicing and premature termination, and identification of hard-to-isolate antigens.
The procedure is expedited by shortening the blocking time and bypassing the secondary antibody-binding steps. This approach allows for fewer opportunities for human error, while maintaining comparable sensitivity to the standard 4.5 hour procedure. Results are highly reproducible with low background; kit allows for detection of nanograms of antigen while leaving quantities of primary antibody of up to 2µg per lane undetected. Procedure does not require secondary antibody, labelling of primary antibody, or as much optimization as classical three-step method.
Kits are compatible with immunoprecipitation methods incorporating proteins A and G, A/G resin, or any secondary antibody resin.
