Ficoll PM400 centrifugation medium is designed for producing density gradients for separation of cells and subcellular components by centrifugation or by sedimentation at unit gravity. It is composed of a neutral, highly-branched, hydrophilic polymer of sucrose that dissolves readily in aqueous solution. Concentrations of up to 50% (w/v) covering densities of up to 1.2g/mL can be achieved. This media features better osmotic properties than sucrose, preserves functional and morphological integrity, and is useful for separating cells that are sensitive to centrifugation, as well as for separating cells of similar density but different size (under conditions of sedimentation at unit gravity). It does not penetrate biological membranes. Medium is in the form of dry powder and serves as the raw material for preparation of Ficoll-Paque gradients. It is also used in applications such as electrophoresis, hybridization, cryopreservation, and as a hapten carrier.

Ficoll-Paque reagents are designed to prepare human mononuclear cells from peripheral blood, bone marrow, and umbilical cord blood via density gradient centrifugation. These sterile density media have established a proven record of isolating high yields of mononuclear cells. Both Ficoll-Paque PLUS and PREMIUM media exhibit consistently low endotoxin levels (<0.12EU/mL).
