Developed for the toughest cloning and protein expression jobs, E. cloni competent cells are optimized to give high yields of recombinant plasmid, cosmid, or BAC DNAs. These cells share the most useful genetic elements of standard cloning and expression strains like DH5alpha*, DH10B*, or BL21 (DE3), and can directly replace them in cloning and expression protocols. However, E. cloni cells feature technology that increases transformation efficiency and reliability, while decreasing costs.

10G SUPREME electrocompetent cells are designed for the most demanding cloning projects or for creating large, complex libraries. 10G ELITE electrocompetent cells are meant to be used with difficult-to-clone DNA or library construction. 10G CLASSIC electrocompetent cells are ideal for general high efficiency cloning. 10G and 10GF' chemically competent cells are meant for rountine cloning and subcloning. All of the 10G cells are phage resistant and can be used to clone cosmids, BAC, and methylated DNA. They can also be used in blue/white screening protocols without IPTG induction.

10GF' ELITE electrocompetent cells are designed for ssDNA production or general high efficiency cloning of methylated DNA. They can also be used in blue/white screening protocols; however, unlike the 10G cells, the 10GF' cells require IPTG induction. These cells are not appropriate for use in BAC or cosmid cloning.

TG1 electrocompetent cells are designed for phage display protein expression, and are also suitable for M13 phage work, general cloning, and blue/white screening. The 5-Alpha chemically competent cells are versatile enough for use in a wide variety of applications, such as routine cloning, subcloning, and plasmid isolation with or without blue/white screening.

EXPRESS BL21 (DE3) electrocompetent cells are designed to enable researchers to clone and express proteins in the same cell. They can be used with cosmid, BAC, and methylated DNA to construct complex expression libraries. EXPRESS BL21 (DE3) chemically competent cells are meant to be used in routine protein expression protocols. These can also be used with cosmid, BAC, and methylated DNA. Both EXPRESS electrocompetent and chemically competent cells are also available with a chloramphenicol-resistant plasmid that encodes T7 lysozyme, which is a natural inhibitor of T7 RNA polymerase. Cells containing pLysS produce a small amount of lysozyme, while the pLysE cells produce a larger amount of lysozyme. The EXPRESS line of cells is not appropriate for use with blue/white screening protocols.

The OverExpressâ„¢ line of cells are E. coli strains that are effective in expressing toxic proteins from all classes of organisms. They contain genetic mutations phenotypically selected for conferring tolerance to toxic proteins. The C41(DE3) strain was derived from BL21(DE3) and has at least one uncharacterized mutation to prevent cell death associated with expression of many recombinant toxic proteins. The C43(DE3) was derived from C41(DE3) by selecting for resistance to a different toxic protein to enable the C43(DE3) cells to express a different set of toxic proteins. As in standard BL21(DE3) strains, pLysS cells produce a small amount of lysozyme. OverExpress cells are also deficient in the Ion and ompT proteases. OverExpress strains can be differentiated from each other and from BL21(DE3) by transformation with the plasmid verification vector pAVD10. This vector is lethal to BL21(DE3) and to induced C41(DE3), but tolerated by C43(DE3), regardless of induction. The vector is supplied with all OverExpress cells.
