GC Cloning is analogous to TA cloning and is based on the discovery that non-proofreading DNA polymerases, such as Taq, add a single, non-templated 3'-G overhang to PCR† products or to blunt DNAs. These kits allow for fast and easy cloning of PCR products up to 10kb in size. The kits are compatible with products produced from either proofreading or non-proofreading DNA polymerases. The pSMART* GC and pGC* Blue vectors each contain complementary single 3'-C overhangs for fast and highly efficient ligation of G-tailed inserts. In addition, since these vectors feature cloning efficiencies >99%, essentially every colony is recombinant. Kits allow up to nanogram amounts of target DNA to be cloned efficiently and without loss of sample.

GC Cloning vectors also incorporate CloneSmart* transcription terminators for increased insert stability when cloning sequences that may be toxic to the host. In addition, the vectors are kanamycin-resistant to eliminate non-transformed satellite feeder colonies that are inherent in ampicillin-resistant TOPO*/TA* vectors. Vectors are available in either a high copy (HK) version, which yields ~300 copies/cell for cloning routine PCR products, or a low copy (LK) version, which yields ~20-50 copies/cell for cloning particularly difficult inserts or DNAs larger than 8kb. The pGC Blue vector includes the option of blue/white screening in the cloning experiment, use the pGC Blue vector. These vectors also contain opposing SP6 and T7 promoters to allow for flexible expression of PCR products.

Kits include E. cloni* 10G competent cells, which are designed for high efficiency cloning. Kits are available with either electrocompetent or chemically competent cells. Choose SUPREME (>=4x1010cfu/µg) or ELITE (>=2x1010cfu/µg electrocompetent cells when cloning large or difficult DNAs or when maximum yields are desired. Choose chemically competent cells (>=1x10⁸cfu/µg) for routine cloning.
