Designed for PCR†and cDNA cloning, as well as shotgun sequencing. The pEZSeq kits are optimized for the highest efficiency of blunt end cloning and sequencing using a blue/white vector. Each pEZSeq vector yields 99% recombinant clones with blunt, dephosphorylated ends, compared to common pUC based vectors that are only 40-70% efficient. The low background of blue colonies from pEZSeq vectors virtually eliminates robotic picking errors and greatly simplifies manual clone selection. The low background also allows transformants to be plated without blue/white screening in cDNA and shotgun library construction.
pEZSeq vectors are designed with strong transcriptional terminators on either side of the lacZ gene to reduce bias against toxic inserts. They also feature a minimal length of DNA between the cloning site and the primer binding sites, enabling the resulting sequence to contain 5-10% more insert sequence. The additional data can reduce the number of clones needed to assemble a contig and can add millions of extra bases to large-scale projects. In addition, the micro vector design minimizes unwanted transposition events, facilitates the cloning of larger inserts, and simplifies subsequent manipulations.
The lac promoter of pEZSeq vectors are constitutively active in E. cloni® 10G cells, so IPTG is not required for the blue/white screen. The lacq allele on the F' of 10GF' efficiently represses the lac promoter, allowing the user the option of adding IPTG to use the blue/white screen to assess cloning efficiency or omitting IPTG induction to reduce any cloning bias that may be created by high expression of the inserts.
Note: Kits with electrocompetent cells are recommended for use with Eppendorf*, Bio-Rad*, and BTX* electroporation cuvettes. They are not recommended for use with Invitrogen* cuvettes.
