The pJW168 vector is designed for in vivo insertion/removal of a DNA sequence through cre-lox site-specific recombination. The vector contains the cre-recombinase gene under the control of the IPTG-inducible lacUV5 promoter, as well as unique restriction sites for cloning and the gene for ampicillin selection. This vector allows DNA fragments to be integrated and then completely removed from the bacterial chromosome. Following recombination, growing transformed bacterial cultures at 42°C (108°F) will efficiently eliminate pJW168 and cease the production of cre recombinase.
The plnvRecA vector is designed to enable P1 transduction in any E.coli strain, making it highly useful in generating mutant strains for microbial genomic and proteomic studies, or for moving genes from one strain to another. This vector can be used in any E. coli strain, unlike other P1 transduction strategies, which can only be used with wild-type (RecA+) E. coli strains.
