Kits measure the activation of small GTPases by isolating them via their specific downstream effectors. The respective binding domain of the downstream effector for each small GTPase is expressed as a GST-fusion protein that is used to pull down the active GTPase. Once the active GTPase is pulled down it can be detected by a Western blot using the specific antibody included in each kit. The assays provide consistent results with less sample because the cell lysate is incubated with both GST-fusion protein and a premeasured SwellGel* immobilized glutathione disc. This also means that there is no need to express and purify GST-PBD or GST-RBD fusion proteins, making the assays quick and easy to run. In addition, kits use spin columns to allow for efficient recovery of the sample.
