When proteins are expressed recombinantly, additional amino acids called fusion tags are often added to aid in purification and manipulation. These kits are available with six histidine residues (6xHis-tagged) or glutathione s-transferase (GST-tagged) fusion proteins. Kits are optimized to extract and purify the highest quality possible in the least amount of time. In addition, the protocols, do not require any manual or mechanical lysing steps. Instead kits include the appropriate cell lysis reagents.
Histidine-tagged recombinant proteins are easily processed because the tag binds to several metals, including nickel, cobalt, and copper. The agarose supports included in the histidine kits are derivatized with nickel chelating groups and iminodiacetic acid. This allows for a tighter bond with the histidine-tagged protein. GST-tagged recombinant proteins show more specific binding than those tagged with histidine, resulting in greater purity in the final samples. Because binding is so specific, the protocol requires the addition of reduced glutathione (included) to a physiologic buffer in order to release the GST fusion proteins from the immobilized glutathione substrates.
