Use the biotin random prime DNA labeling kit (PI17075) to create biotin-labeled probes for Southern blotting. The probes are created when random heptanucleotides containing all possible sequences are annealed to a denatured DNA template. These then act as primers for complementary strand synthesis by DNA polymerase. The product formulation uses biotinylated dCTP instead of dUTP to maximize the streptavidin-HRP binding and increase the sensitivity. Reactions can be done with as little as 100ng of starting DNA template. The Klenow fragment in the labeling kit is exonuclease-free to produce higher yields. Each reaction will yield enough probe sample for three 10x10cm (315/16x315/16") blots.
Once the labeled probes are ready for the membrane, use the chemiluminescent hybridization and detection kit (PI17097) to detect the streptavidin-HRP labeled probe. The kit combines an enhanced luminol substrate for horseradish peroxidase (HRP) with pre-mixed hybridization and blocking buffers to ensure consistent results with sensitivity equal to or greater than radioactive phosphorus techniques. The protocol is amenable to many formats and results are easily recorded by a 1-5 minute exposure to X-ray film or with a CCD camera.
Ordering Information: The biotin random prime DNA labeling kit (PI17075) comes with 100µL 5X heptanucleotide mix, 80µL 5X deoxynucleotide mix, 50µL 10X reaction buffer, 10µL Klenow DNA polymerase, 5µL control DNA, 1mL 5M ammonium acetate, 1mL 0.5M EDTA at pH 8.0, 1mL nuclease-free water, and 20µL biotin-II-dUTP. Reagents included are sufficient for 10 reactions. The chemiluminscence hybridization and detection kit (PI17097) comes with 1.5mL stabilized conjugated streptavidin-HRP, 80mL luminol/enhancer solution, 80mL stable peroxide solution, 125mL hybridization buffer, 375mL 2X hybridization stringency wash buffer, 500mL blocking buffer, 500mL 4X wash buffer, and 500mL substrate equilibration buffer. Reagents provided are sufficient for 1000cm2 of blotting membrane. The complete labeleing and detection kit (PI17175) contains everything found in kits PI17075 and PI17097. The chemiluminescent substrate (PI17295) is available separately. The substrate comes with 50mL each of luminol/enhancer solution and stable peroxide solution and is sufficient for 1000cm2 of blotting membrane.
