By simply measuring the Å280 and Å354 of a modified protein, the concentration and number of biotins incorporated per protein can be determined directly. No HABA or fluro-reporter assay needed. Only a 20µg aliquot of biotin-modified protein in a 100µL volume of buffer is required to yield reproducible results using a standard microplate reader or microcuvette.

The advanced protein biotinylation reagent features a water-soluble sulfo-succinimidyl ester functional group that modifies protein lysine residues in an aqueous buffer system. The linker also contains a UV-traceable bis-aryl hydrazone chromophore that absorbs at 354nm. This UV-signature permits rapid quantification of incorporated biotin by means of nondestructive, two-wavelength spectrophotometric measurement. The linker possesses an extended PEG3 spacer which helps preserve and maintain streptavidin/biotin affinity while enhancing antibody solubility.

The kit includes all the necessary reagents for incorporating traceable biotins on any amine-containing protein. Enough reagents are included for 25µg to 1mg of antibody per labeling with five labeling runs. The protocol is included on an accompanying flash drive. (SoluLink B-9007-105K)
